It has been demonstrated that opiate drugs produce, in the brain, a neural response pattern similar to that described for emotional stimuli. However, the possible relationship between the endorphin system and long-lasting effects such as drug addiction is unknown. To study the possible role of the endogenous opioid system in addictive behaviour, the expression of the proto-oncogene c-fos was evaluated in the CA1, CA3 and dentate gyrus sub-regions of the hippocampal formation after a 24 h stimulation of brain opioid receptors by the opioid agonist DAGO (D-Ala2, D-Leu5-enkephalin), in two groups of Sprague-Dawley rats: the control group (n = 8) was infused with vehicle and the experimental group (n = 8) with the opioid agonist DAGO. The animals were perfused, after the DAGO infusion, and the immunohistochemical evaluation of c-fos expression was performed in the three sub-regions of the hippocampus. No differences in the basal expression of c-fos were found between control and experimental animals. However, a significant increase in c-fos expression was found in the experimental group as compared to control group. The increase in c-fos expression was significantly higher in the dentate gyrus than in the CA1 and CA3 sub-regions. These results suggest that stimulation of brain opioid receptors induces a long-lasting increase in the expression of c-fos in hippocampal neurons.
